Alocrest™ Alocrest™ (vinorelbine liposomes injection, OPTISOME™)

A Novel Targeted Anti-Cancer Compound for Breast and Lung Cancer.

Alocrest™ has completed a Phase I dose-escalation trial demonstrating promising anti-cancer activity as well as acceptable and predictable toxicity:
  • A 47% disease control rate (1PR, 13 SD) across a broad range of doses in mostly heavily pretreated patients with relapsed and/or refractory solid tumors
  • Maximum tolerated dose of 28 mg/m2in heavily pretreated patients that is comparable to that of un-encapsulated vinorelbine. Possibility to increase this dose level in future trials in non-heavily pretreated patients
  • Reversible neutropenia without fever was the most common dose limiting toxicity
  • No peripheral vein irritation

Preclinical data demonstrate the value of Alocrest™ over standard vinorelbine (VRL):
  • Prolonged circulation and 100-fold increased plasma area under concentration-time curve (AUC)
  • 9.5-fold increased VRL exposure (tissue AUC) at tumor sites
  • Up to a 3.5-fold increased therapeutic index in xenograft models

Alocrest has the potential to deliver improved safety and efficacy compared to un-encapsulated vinorelbine injection in target populations as non-small cell lung cancer in the elderly.
